制备高效价的幽门螺杆菌尿素酶B(Ure B)亚单位重组蛋白免疫血清,探讨不同剂量、不同途径对家兔免疫效果的影响。采用0.5,1,5 mg/m L Ure B抗原分别通过皮内(sc)、肌肉(im)、腹腔(ip)3种途径对新西兰大白兔进行免疫,免疫双扩散法测定抗血清效价。结果显示,1 mg/m L抗原剂量免疫效果最佳,优于0.5 mg/m L的剂量,5 mg/m L的抗原剂量造成家兔免疫耐受;皮内注射途径所制备的兔抗血清抗体效价最高。本研究成功制备Ure B重组蛋白多抗血清,同时还优化得到Ure B重组蛋白多抗血清制备的最佳免疫剂量和免疫途径,为今后幽门螺杆菌疫苗的活性测定奠定了基础。
Preparation of high titer of Helicobacter pylori urease B (Ure B) recombinant protein immune serum to explore different doses and different ways on the immune effect of rabbits. New Zealand white rabbits were immunized with 0.5, 1, 5 mg / mL Ure B antigens by intramuscular (sc), intramuscular (im) and intraperitoneal (ip) respectively. The antiserum titers were measured by double immunodiffusion. The results showed that 1 mg / m L antigen dose had the best immunization effect, better than 0.5 mg / m L dose and 5 mg / m L antigen dose resulted in immune tolerance in rabbits. Rabbit antisera Antibody titers highest. In this study, Ure B recombinant polyclonal antiserum was successfully prepared, and the optimal immunization dose and immunization route of Ure B recombinant polyclonal antiserum were optimized, which laid the foundation for the determination of the activity of Helicobacter pylori vaccine in the future.
