目的研究Tesmin基因的表达特点。方法将4d、9d、18d、35d、54d和6月龄小鼠睾丸组织提取RNA,通过RT-PCR分析差异表达基因Tesmin在小鼠睾丸不同发育阶段中的表达。结果基因芯片结果显示4d、9d、18d、35d、54d和6月龄小鼠睾丸中分别是31.4(A)、34(A),1292(P)、865.6(P)、977.2(P)和830.7(P),代表Tesmin基因在4d,9d小鼠中无表达,1 8d龄后开始高表达,RT-PCR结果表明小鼠Tesmin基因在小鼠9d龄及之前的睾丸中没有表达,在18d龄后睾丸中开始高表达,与基因芯片分析结果相一致。结论Tesmin基因为小鼠年龄依赖性表达基因,小鼠Tesmin基因的表达与小鼠精子发生有很强的一致性,推测该基因可能在精子发生中起重要作用。
Objective To study the expression of Tesmin gene. Methods RNA was extracted from testes of 4d, 9d, 18d, 35d, 54d and 6-month-old mice and the expression of differentially expressed gene Tesmin was detected by RT-PCR in different developmental stages of mice testes. Results The result of gene chip showed that the testis in testes of mice on day 4, 9, 18, 35, 54 and 6 were 31.4 (A), 34 (A), 1292 (P), 865.6 (P), 977.2 (P) .There was no expression of Tesmin gene in 4d and 9d mice, and high expression after 18d. RT-PCR results showed that mouse Tesmin gene was not expressed in testes of 9d and before mice, After testicular high expression began, consistent with the results of gene chip analysis. Conclusion Tesmin gene is age-dependent gene expression in mice. The expression of mouse Tesmin gene is highly consistent with that of mouse spermatogenesis. It is speculated that this gene may play an important role in spermatogenesis.
