我们在GM—CFU传统体外琼脂培养(简称全量法)基础上,参照葛忠良等建立的GM—CFC体外微量法(筒称微量法),对“正常人”骨髓及3.5~5个月胎龄的胎肝细胞悬液各9例进行了GM—CFU体外微量琼脂培养。“正常人”骨髓取自本院胸科手术中切除的肋骨,半小时内将骨髓细胞挤压在RPMI1640液,用吸管打散,过7号及41/2号针头后以2000r/min离心10min。弃上清,加适量RPMI1640液打散,淋巴细胞分层液(比重1.077)4ml的离心管内加上骨髓细胞分层以1700r/min离心25min.
