目的 :建立 1 甲基 4苯基吡啶离子 (1 methyl 4 phenylpyridiniumion ,MPP+)诱导的大鼠小脑颗粒细胞凋亡模型 ,探讨肉苁蓉成分是否对MPP+诱导的大鼠小脑颗粒细胞凋亡有保护作用。方法 :用肉苁蓉成分预先孵育大鼠小脑颗粒细胞 ,经MPP+处理这些细胞后 ,用甲基绿 派诺宁染色、DNA凝胶电泳、流式细胞术检测。结果 :浓度为 5 0 μmol·L-1MPP+使小脑颗粒细胞发生典型的凋亡 ,2 5mg·L-1campneosideⅡ具有抗凋亡作用。结论 :以MPP+为诱导剂建立的大鼠小脑颗粒细胞凋亡模型 ,可用于研究与帕金森病 (Parkinson’sdisease ,PD)有关的细胞凋亡的调控机制和筛选抗PD药物 ,中药肉苁蓉成分campneosideⅡ对神经毒素MPP+诱发细胞凋亡具有明显的保护作用
Objective : To establish a model of rat cerebellar granule cell apoptosis induced by 1 methyl 4 phenylpyridinium ion (MPP+), and to investigate whether the composition of Cistanche deserticola has a protective effect on the apoptosis of rat cerebellar granule cells induced by MPP+. METHODS: Rat cerebellar granule cells were pre-incubated with Cistanche, and these cells were treated with MPP+ and then detected with methyl green nononin staining, DNA gel electrophoresis, and flow cytometry. RESULTS: Apoptosis of cerebellar granule cells was observed at a concentration of 50 μmol·L-1 MPP+, and 25 mg·L-1 campneoside II had anti-apoptosis effect. Conclusion: Apoptosis model of rat cerebellar granule cells established with MPP+ inducer can be used to study the regulation mechanism of apoptosis related to Parkinson’s disease (PD) and screening anti-PD drugs.
