为了攻克利用DNA指纹技术进行玉米种子纯度鉴定中存在的技术难关,对玉米单株幼芽DNA快速提取 方法进行了研究,结果表明:利用新方法提取DNA,不用液氮、不用研磨、不用离心、不用沉淀,不用SDS、不用CTAB、 不用氯仿,并且提取的DNA不降解、完全可以满足利用SSR分子标记进行DNA指纹分析的需要。一个人提取100 株幼芽(1个检测样品)DNA只需30 min左右,1 d可以提取2000株幼芽(20个检测样品)的DNA,为DNA指纹技术 在玉米种子纯度及真伪快速鉴定中的广泛应用解决了关键性技术难题。
In order to overcome the technical difficulties in the identification of maize seed purity by DNA fingerprinting, the rapid DNA extraction method of maize single shoots was studied. The results showed that DNA was extracted by new method without liquid nitrogen, without grinding, without centrifugation, No precipitation, no SDS, no CTAB, no chloroform, and the extracted DNA does not degrade, completely meeting the need of using SSR molecular markers for DNA fingerprinting. One person can extract DNA of 100 sprouts (1 test sample) in about 30 minutes and extract DNA of 2000 sprouts (20 test samples) on 1 day, which is a rapid identification of purity and authenticity of corn seeds by DNA fingerprinting The wide range of applications addresses key technical challenges.
