目的通过流式细胞技术分选人舌癌细胞株Tca8113中的侧群细胞(SP细胞)、非侧群细胞(MP细胞),检查ABCG2蛋白在SP细胞、MP细胞、T(Tca8113)细胞中的表达情况,并探讨其意义。方法用流式细胞仪检测SP细胞、MP细胞、未分选Tca8113细胞中的ABCG2表达情况。结果人舌癌细胞株Tca8113中存在SP细胞,ABCG2在Tca8113SP细胞中较高表达(表达率13.16%),在MP细胞中几乎不表达(表达率1.07%)、在Tca8113细胞中少量表达(表达率3.29%)。结论人舌癌细胞株Tca8113中的SP细胞较高表达ABCG2,通过ABCG2+分选侧群细胞,将有可能进一步分选纯化类肿瘤干细胞,为今后分离肿瘤干细胞提供有效手段。
OBJECTIVE: To sort side population cells (SP cells) and non-side population cells (MP cells) in human tongue cancer cell line Tca8113 by flow cytometry to examine the effect of ABCG2 protein in SP cells, MP cells and T (Tca8113) cells Express the situation and explore its meaning. Methods Flow cytometry was used to detect ABCG2 expression in SP cells, MP cells and untreated Tca8113 cells. Results There were SP cells in human tongue cancer cell line Tca8113. ABCG2 was highly expressed in Tca8113SP cells (13.16%), almost not expressed in MP cells (1.07%), and slightly expressed in Tca8113 cells 3.29%). Conclusion The human tongue cancer cell line Tca8113 expressed high levels of ABCG2 in the SP cells. Sorting the side population cells by ABCG2 + might further purify the tumor-like stem cells and provide an effective method for the isolation of cancer stem cells in the future.
