药物动力学研究中应用统计矩理论分析药物浓度—时间曲线(c-t曲线)的方法已被广泛地采用。这种方法基于以下假设:血浆药物浓度的经时过程是一统计分布曲线。但实际上它并不是统计分布曲线。因此,用这种方法估算出的药物在体内平均滞留时间(MRT)只有当c-t曲线呈单指数方程时才是真实的MRT,在其他情况下,能引起系误统差。作者讨论了引入误差的原因,并给出一种新的方法,求算瞬时注入给药时的MRT。药动学中常用的统计矩可简单地表示如下:
Pharmacokinetic studies using statistical moment theory to analyze drug concentration-time curves (c-t curves) have been widely adopted. This method is based on the assumption that the time course of plasma drug concentration is a statistical distribution curve. But in fact it is not a statistical distribution curve. Therefore, the MRT estimated in this way in vivo is the true MRT only if the c-t curve is in the form of a single exponential equation, and in other cases it can cause a systematic error. The authors discuss the reasons for the introduction of errors and propose a new method to calculate MRTs for instantaneous administration.
