目的 探讨胰腺癌细胞Flt- 1受体表达与p5 3、ras基因突变之间的关系,了解Flt- 1受体表达与p5 3、ras基因突变在胰腺肿瘤血管形成中的作用。方法 采用免疫组织化学EnVisionTM方法检测40例原发性胰腺癌组织Flt- 1受体和p5 3、ras基因的表达;采用χ2 检验分析Flt- 1受体表达与p5 3、ras突变基因的相关性。结果 Flt- 1受体和p5 3、ras基因的阳性表达率分别为77.5 %(3 1/4 0 )和5 0 % (2 0 /4 0 )、60 % (2 4/4 0 )。经连续切片对比分析表明,在Flt 1受体表达较高的区域,p5 3、ras基因的表达亦较强,Flt 1受体表达与p5 3基因突变呈显著相关性(χ2 =4.3 5 ,P <0 .0 5 ) ,但与ras基因表达之间无显著相关性(χ2 =2 .12 ,P >0 .0 5 )。结论 胰腺癌细胞Flt 1受体表达与p5 3基因突变之间有显著相关性,与ras基因突变之间无显著相关性,提示野生型p5 3基因发生突变可能会失去抑制Flt 1受体表达的功能,从而导致胰腺肿瘤血管的新生,促进肿瘤细胞生长;ras突变基因可能通过其他的途径参与胰腺肿瘤新生血管的形成。
Objective To investigate the relationship between the expression of Flt-1 receptor and the mutation of p5 3 and ras in pancreatic cancer cells, and to investigate the role of Flt-1 receptor and mutation of p5 3 and ras in angiogenesis of pancreatic tumor. Methods EnVisionTM immunohistochemistry was used to detect the expression of Flt-1 receptor and p5 3, ras gene in 40 cases of primary pancreatic cancer. The correlation between Flt-1 receptor expression and the mutation of p5 3 and ras was analyzed byχ2 test . Results The positive rates of Flt-1 receptor, p5 3 and ras gene were 77.5% (3 1/4 0), 50% (20/40) and 60% (24/40) respectively. The results of serial sections showed that there was a strong expression of p5 3 and ras genes in the regions with high expression of Flt 1 receptor, and the expression of Flt 1 receptor was significantly correlated with the mutation of p5 3 gene (χ2 = 4.3 5, P <0.05), but no significant correlation with ras gene expression (χ2 = 2.12, P> 0.05). Conclusion There is a significant correlation between the expression of Flt 1 receptor and the mutation of p5 3 gene in pancreatic cancer cells. There is no significant correlation between the expression of Flt 1 gene and the mutation of ras gene, which indicates that the mutation of wild type p5 3 gene may lose the expression of Flt 1 receptor Function, leading to pancreatic tumor angiogenesis and promote tumor cell growth; ras mutant genes may be involved in the formation of pancreatic neoplasms by other means.
