体外培养胎儿心肌细胞,待细胞贴壁成层时接种CoxsackieB3病毒(CVB3),24小时后收集细胞。应用电镜、DNA凝胶电泳分析及原位末端标记法(TUNEL)检测发现,感染病毒心肌细胞每视野均有70%以上呈阳性染色,DNA凝胶电泳出现“梯状带”;对照细胞呈现阳性染色者每视野不超过5%,DNA凝胶电泳只见一条正常基因组DNA带。实验结果提示,CVB3可诱导体外培养人心肌细胞的凋亡。
Fetal cardiomyocytes were cultured in vitro. Coxsackie B3 virus (CVB3) was inoculated when the cells adhered to each other and cells were harvested 24 hours later. Electron microscopy, DNA gel electrophoresis analysis and TUNEL assay showed that more than 70% of infected myocardium were stained positive in each field of view, and “ladder-like band” appeared in DNA gel electrophoresis. Control cells showed positive staining Dyeing per field does not exceed 5%, DNA gel electrophoresis saw only a normal genomic DNA band. Experimental results suggest that CVB3 can induce apoptosis in cultured human cardiomyocytes in vitro.
