目的研究前列腺癌中脂肪酸合成酶(fatty acid synthase,FASE)的表达及其临床意义。方法对68例前列腺癌手术或穿刺活检患者的石蜡标本进行FASE的免疫组织化学染色和半定量分析,并与良性前列腺增生进行对照。结果FASE在良性前列腺增生组织中的阳性表达率(12.8%)明显低于前列腺癌组织(47.1%,P<0.05)。FASE表达与临床分期、病理分级、骨转移有关(P<0.05);与年龄、淋巴结转移、远处转移无关(P>0.05)。FASE阳性和阴性患者的生存率有显著性差异(P=0.002)。结论FASE在前列腺癌组织中表达增高,酶活性增强;FASE在前列腺癌组织中表达增高有提示不良预后的作用。
Objective To study the expression of fatty acid synthase (FASE) in prostate cancer and its clinical significance. Methods Immunohistochemical staining and semi-quantitative analysis of FASE were performed on paraffin specimens from 68 patients with prostate cancer or biopsy, and compared with benign prostatic hyperplasia. Results The positive rate of FASE in benign prostatic hyperplasia (12.8%) was significantly lower than that in prostatic carcinoma (47.1%, P <0.05). FASE expression was correlated with clinical stage, pathological grade and bone metastasis (P <0.05), but not with age, lymph node metastasis and distant metastasis (P> 0.05). There was a significant difference in survival between FASE positive and negative patients (P = 0.002). Conclusions The expression of FASE is increased in prostate cancer tissue and the activity of FASE is increased. The increased expression of FASE in prostate cancer tissue suggests the role of FASE in poor prognosis.
