目的探讨基质金属蛋白酶1(MMP-1)、基质金属蛋白酶7(MMP-7)基因在哈萨克族食管癌和正常组织的表达及与病理特征关系,分析两者表达的相关性。方法采用逆转录聚合酶链反应(RT-PCR)方法在45例哈萨克族食管癌癌组织及其远端正常组织中检测MMP-1与MMP-7基因的表达。结果在45例食管癌组织中,MMP-1、MMP-7基因的表达阳性率分别为75.56%和73.33%,在远端正常组织的表达阳性率分别为15.56%和31.11%,二者差异均有统计学意义(P<0.05);MMP-1表达与TNM分期有关,MMP-7表达与TNM分期及淋巴结转移有关(P<0.05);MMP-1与MMP-7基因的表达具有正相关性(P<0.05)。结论 MMP-1和MMP-7在哈萨克族食管癌发生发展中起一定作用,两者之间具有协同作用,促进食管癌发病进程。
Objective To investigate the expression of matrix metalloproteinase-1 (MMP-1), matrix metalloproteinase-7 (MMP-7) gene in esophageal cancer and normal tissues of Kazak and their relationship with pathological features, and to analyze their correlation. Methods Reverse transcription polymerase chain reaction (RT-PCR) was used to detect the expression of MMP-1 and MMP-7 in 45 Kazakh esophageal cancer tissues and their distant normal tissues. Results The positive expression rates of MMP-1 and MMP-7 in 45 esophageal cancer tissues were 75.56% and 73.33%, respectively, and the positive rates in distal normal tissues were 15.56% and 31.11%, respectively (P <0.05). The expression of MMP-1 was related to TNM staging. The expression of MMP-7 was correlated with TNM staging and lymph node metastasis (P <0.05). There was a positive correlation between the expression of MMP-1 and MMP-7 (P <0.05). Conclusions MMP-1 and MMP-7 play a role in the carcinogenesis and progression of esophageal cancer in Kazakh. The two have a synergistic effect to promote the progression of esophageal cancer.
