目的考察不同粒径的藏茜草粉体中大叶茜草素含量变化特征和规律。方法制备10~300目的藏茜草粉样品,采用高效液相色谱法测定大叶茜草素含量,色谱柱为Kromasil C18(4.6mm×250mm),流动相甲醇-水-四氢呋喃(343∶90∶3),检测波长250nm。结果大叶茜草素对照品在0.52~2.60μg范围内,进样量与峰面积呈良好线性关系,回归方程为Y=306042.1X+103512.3,r=0.9993,平均回收率为99.5%,RSD为2.61%,具有较好的精密度和重复性。结论大叶茜草素的含量随着藏茜草粉粒径的变小而增加,当粉碎到80目以上时,大叶茜草素的含量不再随粒径的减小而增加。
OBJECTIVE To investigate the characteristics and regularities of the changes of magnolia macrophylla in different sizes of Tibetan madder meal. METHODS: The samples of Tibetan madder meal with the size of 10-300 were prepared and the contents of the main fractions of raneopalmarin were determined by HPLC. The mobile phase consisted of Kromasil C18 (4.6 mm × 250 mm), mobile phase of methanol-water-tetrahydrofuran (343: 90: 3) , Detection wavelength 250nm. The results showed that there was a good linear relationship between the injection volume and peak area in the range of 0.52 ~ 2.60μg. The regression equation was Y = 306042.1X + 103512.3, r = 0.9993, the average recovery was 99.5%, RSD was 2.61 %, With better precision and repeatability. Conclusion The content of magnolia extract increased with the decrease of particle size of Tibetan madder meal. When smashed to more than 80 meshes, the content of magnolia extract did not increase with the decrease of particle size.
