目的:探索鲜壁虎粗提物经蛋白酶酶解后对小鼠H22移植瘤的抑制作用。方法:采用可溶性氮含量(SNTCA指数)法进行酶解度测定,鲜壁虎粗提物经胃蛋白酶、木瓜蛋白酶组合水解得到酶解物;采用小鼠H22肝癌移植模型,分阴性对照组、阳性对照组、粗提物组及粗提物酶解组,测小鼠瘤重及脾和胸腺指数。结果:粗提物组、粗提物酶解组对H22荷瘤小鼠肿瘤抑制率分别为29.17%、48.99%,粗提酶解组与阴性对照组相比有显著差异(P<0.05)。粗提物组及粗提物酶解组的荷瘤小鼠脾、胸腺质量及其指数与阴性对照组相比无差异。结论:鲜壁虎粗提物组及粗提物酶解组对小鼠H22移植瘤具有抑制作用,酶解后增加了鲜壁虎粗提物的抗肿瘤活性。
Objective: To explore the inhibitory effect of protease on the H22 xenografts of mice after the crude extract of Gekko gentianus. Methods: The content of soluble nitrogen (SNTCA index) was used to determine the degree of hydrolysis. The crude extract of Gekko gecko was hydrolyzed by pepsin and papain to obtain the hydrolyzate. H22 hepatocellular carcinoma model was used in the mice. The negative control group, positive control Group, crude extract group and crude extract enzymatic group, measured tumor weight and spleen and thymus index. Results: The tumor inhibition rates of crude extract group and crude extract enzymatic hydrolysis group were 29.17% and 48.99% respectively in H22 tumor-bearing mice. There was a significant difference (P <0.05) between the crude extract group and the negative control group. The crude extract group and crude extract enzymatic group of tumor-bearing mice spleen, thymus quality and index no difference compared with the negative control group. Conclusion: The crude gecko crude extract group and the crude enzyme hydrolyzate group can inhibit the growth of mouse H22 xenografts, and the enzymatic hydrolysis increases the antitumor activity of the crude gecko crude extract.
