目的:以CC l4损伤肝纤维化大鼠作为模型,用正交设计方法,对文献报道有抗肝纤维化作用的中药丹参、黄芪及苦参进行多因素、多水平及交互作用的研究。方法:雄性w istar大鼠48只,随机分为3组,正常对照组(n=8)、模型组(n=8)、实验组(n=32),应用CC l4溶液皮下注射及高脂玉米粉饲料喂养6周,证实造模成功后,采用3因素2水平(取高、低剂量)正交设计,按L8(27)正交表要求将实验组再次随机分为8组(n=4),各实验小组分别以不同实验药物灌胃,6周后观察各组大鼠HA、PⅢP、LN、ⅣC指标,测定肝组织羟脯氨酸(HYP),同时作肝脏病理切片检查。结果:(1)丹参和黄芪能使肝纤维化四项指标显著降低,使肝组织羟脯氨酸含量显著降低(P均<0.05)。(2)丹参和黄芪合用能降低LN和ⅣC,能降低肝组织羟脯氨酸含量(P均<0.05)。(3)丹参、黄芪和苦参合用有交互作用(P均<0.05)。结论:(1)丹参和黄芪单味药有明显的抗肝纤维化作用。(2)丹参、黄芪和苦参合用有交互作用,三药合用效果最佳,最佳组方为第7组,即丹参(高剂量)、黄芪(高剂量)、苦参(低剂量)。
OBJECTIVE: To study the effect of multi-factors, multi-levels and interactions of Salvia Miltiorrhiza, Radix Astragali and Sophora flavescens Ait. on the liver fibrosis in the literature. Methods: Forty male Wistar rats were randomly divided into 3 groups: normal control group (n=8), model group (n=8), experimental group (n=32), subcutaneous injection of CC l4 solution and high fat Cornmeal feed was fed for 6 weeks. After confirming that the model was successful, the 3 factors and 2 levels (high and low doses) were used for orthogonal design, and the experimental group was randomly divided into 8 groups again according to the L8 (27) orthogonal table requirements (n= 4) Each experimental group was given intragastric administration of different experimental drugs. After 6 weeks, the indexes of HA, PIIIP, LN and IVC in each group were observed. Hepatic hydroxyproline (HYP) was measured and hepatic pathological examination was performed. Results: (1) Salvia miltiorrhiza and Radix Astragali could significantly reduce the four indexes of hepatic fibrosis and significantly reduce the content of hydroxyproline in liver tissue (P < 0.05). (2) Salviae miltiorrhizae and Radix Scutellariae can reduce LN and IVC and reduce the content of hydroxyproline in liver tissue (P<0.05). (3) There were interactions between Salvia Miltiorrhizae, Radix Astragali and Sophora flavescens (P <0.05). Conclusion: (1) The single herb of Salvia miltiorrhiza and Radix Astragali has obvious anti-hepatic fibrosis effect. (2) Salvia miltiorrhiza, Astragalus membranaceus and Sophora flavescens combined with interactions, the best combination of three drugs, the best group for the seventh group, that is, Danshen (high dose), Radix (high dose), Sophora (low dose).
