目的观察visfatin对SW872成熟脂肪细胞炎性因子TNF-α、单核细胞趋化蛋白-1(MCP-1)和脂联素蛋白分泌及mRNA表达的影响。方法体外培养SW872细胞,诱导细胞分化,不同浓度visfatin作用于SW872成熟脂肪细胞,孵育24h后收获细胞培养上清和RNA,分别采用ELISA法和RT-PCR法检测上清中TNF-α、MCP-1和脂联素的蛋白分泌及mRNA表达水平。结果与对照组比较,50μg/Lvisfatin即显著促进SW872脂肪细胞TNF-α、MCP-1分泌和mRNA表达及抑制脂联素分泌和mRNA表达;TNF-α、MCP-1和脂联素的蛋白分泌水平均较对照组显著增加[(169.77±13.69)vs(149.30±17.40)ng/g,(1.36±0.14)vs(1.21±1.09)μg/g,(258.53±58.59)vs(330.59±65.00)μg/gPa<0.05],TNF-α和MCP-1mRNA的表达较对照组显著增加[(0.50±0.02)vs(0.45±0.02),(0.55±0.03)vs(0.48±0.01)Pa<0.05],而脂联素mRNA表达较对照组显著下降[(0.90±0.02)vs(0.94±0.04)P<0.05]。当visfatin水平增加至250μg/L时,脂肪细胞TNF-α[(303.20±17.50)ng/g]和MCP-1[(2.00±1.27)μg/g]的蛋白分泌水平较对照组均显著上升(Pa<0.01),而脂联素的蛋白分泌水平[(151.59±36.38)μg/g]则显著减少(P<0.01);同时,TNF-α和MCP-1的mRNA表达水平[(0.82±0.06)ng/g,(0.87±0.06)μg/g]较对照组亦显著增加(Pa<0.01),而脂联素的mRNA表达水平(0.58±0.04)μg/g则显著减少(Pa<0.01)。结论visfatin通过调节促炎性因子和抗炎因子的表达参与了SW872脂肪细胞炎性反应的发生发展。
Objective To observe the effect of visfatin on the secretion and mRNA expression of inflammatory cytokines TNF-α, MCP-1 and adiponectin in SW872 mature adipocytes.
