目的采用改进高效液相色谱法(HPLC)测定心舒丸中丹参酮ⅡA的含量。方法采用Kromasil-C18色谱柱,流动相为甲醇-水(80:20),流速为1.0ml·min-1。结果丹参酮ⅡA在0.10~0.60μg范围内线性关系良好(r=0.9994),回收率为100.10%,RSD为0.58%。改进方法测定丹参酮ⅡA的含量,出峰时间更快,丹参酮ⅡA主峰与其他峰分离度和拖尾因子均符合要求;两种方法测定三批心舒丸中丹参酮ⅡA含量无明显差异。结论改进HPLC法测定丹参药材中丹参酮ⅡA的含量,具有较好的方法重现性,结果准确,可作为丹参酮ⅡA含量测定的替代方法。
Objective To determine the content of tanshinone ⅡA in Xinshu pill by improved high performance liquid chromatography (HPLC). Methods Kromasil-C18 column was used. The mobile phase was methanol-water (80:20) and the flow rate was 1.0 ml · min-1. Results Tanshinone Ⅱ A showed good linearity (r = 0.9994) in the range of 0.10-0.60 μg with a recovery of 100.10% and a RSD of 0.58%. Improved method for the determination of tanshinone Ⅱ A content, peak time faster, tanshinone Ⅱ A main peak and other peak separation and tailing factors are in line with requirements; two methods to determine the three batches of Xinshu pill in tanshinone Ⅱ A content was no significant difference. Conclusion The improved HPLC method for the determination of tanshinone Ⅱ A in Radix Salviae Miltiorrhizae, has good reproducibility and accurate results, and can be used as an alternative method for the determination of tanshinone Ⅱ A.
