目的观察三七总皂苷(Panaxnotoginsengsaponins,PNS)对泡沫细胞ABCA1和LXRαmRNA表达及脂质沉积的影响。方法提取SD大鼠腹腔巨噬细胞纯化后,与高、中、低剂量PNS(80、40、20μg/ml)及氧化低密度脂蛋白共孵育48h。油红O染色观察细胞内脂质堆积情况,酶法测定细胞内总胆固醇(TC)、游离胆固醇(FC)、胆固醇酯(CE)的含量,并计算CE/TC值。采用RT-PCR法检测泡沫细胞ABCA1和LXRαmRNA表达情况。结果模型组细胞中TC、FC、CE含量及CE/TC值均显著高于对照组(P<0.05);与模型组相比,PNS高、中剂量组细胞内TC、FC、CE含量及CE/TC值均显著降低(P<0.05)。PNS高、中剂量组中ABCA1及LXRαmRNA的表达均显著高于模型组(P<0.01)。结论PNS可以上调ABCA1和LXRαmRNA表达并减少巨噬细胞源性泡沫细胞中脂质沉积。
Objective To observe the effects of panaxnotoginsengsaponins (PNS) on the expression of ABCA1 and LXRα mRNA and lipid deposition in foam cells. Methods SD rat peritoneal macrophages were purified and incubated with high, medium and low doses of PNS (80,40,20μg / ml) and oxidized low density lipoprotein for 48h. Oil red O staining was used to observe intracellular lipid accumulation. Enzyme method was used to determine the content of total cholesterol (TC), free cholesterol (FC) and cholesterol ester (CE). The expression of ABCA1 and LXRαmRNA in foam cells was detected by RT-PCR. Results The contents of TC, FC, CE and CE / TC in the model group were significantly higher than those in the control group (P <0.05). Compared with the model group, the contents of TC, FC, CE and CE / TC values were significantly lower (P <0.05). The expression of ABCA1 and LXRαmRNA in PNS high and middle dose groups were significantly higher than those in model group (P <0.01). Conclusion PNS can up-regulate ABCA1 and LXRα mRNA expression and decrease lipid deposition in macrophage-derived foam cells.
