本文研究了大柴胡颗粒(DCHKL)对四氧嘧啶(AXN)致胰岛细胞损伤的保护作用。采用四甲基偶氮唑盐(MTT)法检测大柴胡颗粒对胰岛细胞活性的影响;用ELISA法检测其对胰岛细胞胰岛素基础分泌量和高糖刺激胰岛素分泌量的影响;采用流式细胞仪检测胰岛细胞的凋亡率,同时以RT-PCR检测胰岛细胞的抗凋亡基因(Bcl-2)和促凋亡基因(Bax)的表达。大柴胡颗粒能明显增强胰岛细胞的活性(P<0.01);同时高糖刺激胰岛素分泌量明显上升(P<0.01);能抑制胰岛细胞的凋亡,降低细胞的凋亡率(P<0.01);并且出现上调Bcl-2基因的表达,下调Bax基因表达。结果表明,大柴胡颗粒能提高细胞活性,增加胰岛素分泌,抑制胰岛细胞的凋亡并且上调抗凋亡基因表达,下调促凋亡基因的表达。大柴胡颗粒对胰岛细胞具有一定的保护作用。
In this paper, the protective effect of DCHF on islet cell injury induced by alloxan (AXN) was studied. The effect of Dachaihu granule on islet cell activity was detected by MTT method. The effect of Dachaihu granule on islet cell activity was examined by ELISA. The effect of Dachaihu granule on basal insulin secretion and insulin secretion was examined by ELISA. The apoptosis rate of islet cells was detected. The expressions of anti-apoptotic gene (Bcl-2) and pro-apoptotic gene (Bax) in islet cells were detected by RT-PCR. Dachaihu granules can significantly increase the activity of islet cells (P <0.01); at the same time, high glucose stimulated insulin secretion increased significantly (P <0.01); It can inhibit islet cell apoptosis and reduce cell apoptosis rate (P <0.01) ; And up-regulation of Bcl-2 gene expression, down-regulation of Bax gene expression. The results showed that Dachaihu granules can increase cell viability, increase insulin secretion, inhibit islet cell apoptosis and up-regulate anti-apoptotic gene expression, and down-regulate pro-apoptotic gene expression. Dachaihu particles of islet cells have a protective effect.
