博莱霉素失活酶存在于各种动物组织中并可从小鼠肝脏提取,在小鼠诱发皮肤鳞癌中的含量显然较肉瘤为低。此酶能水解博莱霉素分子中氨基丙氨酸部分的羟酰胺基团而使之失活。本文报导,用亲和层析法提取博莱霉素失活酶。在博莱霉素各组分中,博莱霉素 A_5适宜于亲和层析,因它的末端胺即精脒部分远离羧酰胺基团,用溴化氰激活的方法,使博莱霉素 A_5与珠状琼脂糖凝胶4B(Sepllarose 4B)
Bleomycin-inactivating enzymes are present in various animal tissues and can be extracted from the liver of mice and are clearly lower in sarcoma-induced cancer in mice than sarcomas. This enzyme inactivates the hydroxyamide group of the alanine moiety in the bleomycin molecule. This article reports that bleomycin inactivation enzyme was extracted by affinity chromatography. Among the components of bleomycin, bleomycin A_5 is suitable for affinity chromatography because its terminal amine, ie, the spermine moiety, is far away from the carboxamide group and is activated by cyanogen bromide.
