目的 获得新西兰大白兔角膜缘碱烧伤前后参与修复调控的差异表达基因。 方法 主要采用细胞培养、体外碱烧伤模型的构建、免疫细胞化学、抑制消减杂交和生物信息学等方法。 结果 初步认定样品之一与编码人类血小板反应蛋白 2(TSP2)的基因tsp2高度同源,其中前 698bp序列与人类tsp2基因完全一样,后 230bp与一种人类Trp-4相关蛋白样的新蛋白(人类染色体定位为 20q11. 21-12)相似,后者属于调控蛋白类,具典型的CpG岛特征。目前已在Gen Bank中登录(AF454919)。 结论 该样品可能是TSP家族中TSP2系列的新成员,它对细胞去黏合状态的诱导和抑制血管发生功能的协调可能是促进角膜碱烧伤后创伤愈合的重要原因。
Objective To obtain differentially expressed genes involved in repair regulation in corneal limbal alkali burns in New Zealand white rabbits. The methods mainly used cell culture, construction of in vitro alkali burn model, immunocytochemistry, suppression subtractive hybridization and bioinformatics methods. The results preliminarily confirmed that one of the samples was highly homologous to the gene tsp2 encoding human thrombospondin 2 (TSP2), with the first 698 bp sequence being identical to the human tsp2 gene and the latter 230 bp being identical to a human Trp-4 related protein-like novel protein Human chromosome locates at 20q11. 21-12), which belongs to regulatory proteins with typical CpG island characteristics. Currently registered in Gen Bank (AF454919). Conclusion This sample may be a new member of the TSP2 family in the TSP family. It may play an important role in promoting the wound healing after corneal alkali burn by inducing the cell debonding and inhibiting the angiogenesis.
