植物器官脱落对植物有重要的影响。微管广泛参与植物生长发育、逆境胁迫及植物与环境互作等过程。本文首先建立了棉花叶柄细胞微管免疫荧光组织化学染色法,并利用棉花离体脱落系统进行研究,发现:棉花功能期叶片叶柄非离区厚壁组织细胞微管以斜向排列为主,而离区厚壁组织细胞微管排列方向较为复杂,部分细胞以斜向排列为主,部分细胞则是随机排列。在脱落发生早期,叶柄非离区厚壁组织细胞与离区厚壁组织细胞微管均发生变化,其中非离区厚壁组织细胞主要是排向的变化,变化规律是斜向→纵向→重新斜向;而离区厚壁组织细胞微管则发生可逆性解聚。本研究结果为进一步揭示细胞微管变化与脱落发生的关系奠定了基础。
Plant shedding has a major impact on the plant. Microtubules are widely involved in plant growth and development, stress tolerance, and plant-environment interactions. In this paper, we first established the microtubules of cotton petiole cells by immunofluorescence histochemical staining and the cotton detached shedding system. The results showed that: The arrangement direction of microtubules in thick-walled tissue cells from the area is more complicated. Some cells are mainly arranged in an oblique direction and some cells are randomly arranged. In the early stage of shedding, the thick-walled tissue cells in the non-detached area of petiole and the microtubules of thick-walled tissue cells in the area were changed. The thick-walled tissue cells in the non-exfoliated area mainly changed in the direction of discharge, and the variation was oblique → longitudinal → re- Oblique; and away from the thick-walled tissue microtubules are reversibly depolymerization. The results of this study laid the foundation for further revealing the relationship between the changes of microtubules and the occurrence of shedding.
