目的研究黄体酮对实验性变应性脑脊髓炎(experimental allergic encephalomyelitis,EAE)大鼠的治疗作用。方法采用注射豚鼠脊髓匀浆的方法制作雄性Wistar大鼠EAE模型。当EAE大鼠出现神经症状后,开始在治疗组腹腔内注射溶于2.5ml芝麻油中的黄体酮(4mg·kg-1·d-1,连续注射4d),模型组注射等量的芝麻油。分别对两组大鼠进行运动功能评分和脑组织的电镜检查、LFB染色、锇酸Marchi’s染色。染色结果采用IPP图像分析软件计算光密度(OD)值。结果治疗组和模型组神经功能评分无明显区别,但是在开始治疗后第6、10、25天,治疗组正常髓鞘的积分光密度值明显高于模型组,而退变髓鞘的积分光密度值明显低于模型组。结论黄体酮有利于EAE大鼠中枢神经系统的髓鞘再生,促进髓鞘修复。
Objective To study the therapeutic effect of progesterone on experimental allergic encephalomyelitis (EAE) rats. Methods EAE model of male Wistar rats was made by injecting guinea pig spinal cord homogenate. When EAE rats developed neurological symptoms, progesterone (4 mg · kg-1 · d-1, 4 consecutive days) dissolved in 2.5 ml sesame oil was injected into the treatment group intraperitoneally. The model group was injected with the same amount of sesame oil. Motor function scores and brain tissue electron microscopy, LFB staining and Marchi’s staining of osmium tetroxide were performed on the two groups respectively. Dyeing Results Optical density (OD) values were calculated using IPP image analysis software. Results There was no significant difference between the treatment group and the model group in neurological function score, but at the 6th, 10th, 25th day after the start of treatment, the integral optical density value of the normal myelin sheath in the treatment group was significantly higher than that in the model group, The density value was significantly lower than the model group. Conclusions Progesterone is beneficial to remyelination of central nervous system of EAE rats and promote myelin sheath repair.
