用抗615纯系小鼠的正常肝细胞抗体处理的H_(615)瘤细胞免疫BALB/c系小鼠,取其脾细胞与小鼠骨髓瘤细胞融合,以间接免疫荧光法和细胞ELISA两种方法检测,建立两株分泌抗H_(615)单克隆抗体的杂交瘤细胞系8A58和12C_6。两杂交瘤细胞系所产生的单克隆抗体与正常615系小鼠的肝、脾、肺、肾、脑、心肌、横纹肌、胸腺组织细胞以及小鼠肿瘤细胞、人肝细胞性肝癌细胞均无交叉反应。证明两种单克隆抗体具有一定的特异性。两种单克隆抗体均属IgG_1。
BALB / c mice were immunized with H_ (615) tumor cells treated with normal hepatocyte antibody against 615 pure mice. The spleen cells were fused with mouse myeloma cells. The indirect immunofluorescence and cell ELISA Methods Two hybridoma cell lines 8A58 and 12C_6 secreting anti-H_ (615) monoclonal antibodies were established. Two hybridoma cell lines produced monoclonal antibodies and 615 normal mice liver, spleen, lung, kidney, brain, myocardium, striated muscle, thymic cells, as well as mouse tumor cells, human hepatocellular carcinoma cells without cross reaction. Two kinds of monoclonal antibodies proved to have certain specificity. Both monoclonal antibodies are IgG_1.
