目的探讨转染转移相关基因1(Notch1)和血管内皮生长因子(VEGF)在食管鳞癌组织中的表达情况及临床意义。方法收集食管鳞癌组织标本78例为实验组,并选取癌旁正常食管组织标本42份为对照组,采用EliVision~(TM)免疫组织化学法检测各组Notch1和VEGF的表达情况。Kaplari-Meier法计算患者的3年生存率,Notch1蛋白阳性表达和阴性表达患者的生存差别。结果食管鳞癌组织中Notch1和VEGF表达定位于细胞浆,表达水平与正常食管黏膜组织中的表达差异有统计学意义(P<0.05)。Notch1与肿瘤淋巴结转移、c TNM有关(P<0.05),VEGF的表达与肿瘤淋巴结转移、c TNM、分化程度有关(P<0.05);Notch1和VEGF之间表达呈负相关(P<0.05)。Notch1蛋白阳性表达患者的3年生存率显著低于Notch1蛋白阴性表达者(P<0.05)。结论 Notch1和VEGF在食管鳞癌的表达情况在其发生、发展和转移过程中起到一定的作用,并且其表达水平与预后直接相关,提示二者临床检测可为评估食管鳞癌严重程度提供参考。
Objective To investigate the expression of Notch1 and VEGF in esophageal squamous cell carcinoma and its clinical significance. Methods 78 cases of esophageal squamous cell carcinoma tissue samples were collected as experimental group and 42 cases of adjacent normal esophageal tissue samples were selected as control group. The expression of Notch1 and VEGF in each group was detected by EliVision TM immunohistochemical method. Kaplan-Meier method was used to calculate the 3-year survival rate, survival difference between Notch1 protein positive and negative expression patients. Results The expression of Notch1 and VEGF in esophageal squamous cell carcinoma was localized in the cytoplasm. The expression level of Notch1 and VEGF was significantly different from that in normal esophageal mucosa (P <0.05). Notch1 was correlated with lymph node metastasis and c TNM (P <0.05). The expression of VEGF was correlated with tumor lymph node metastasis, c TNM and differentiation (P <0.05). There was a negative correlation between the expression of Notch1 and VEGF (P <0.05). The 3-year survival rate of Notch1 positive patients was significantly lower than that of Notch1 negative patients (P <0.05). Conclusion The expression of Notch1 and VEGF in esophageal squamous cell carcinoma plays an important role in the occurrence, development and metastasis of esophageal squamous cell carcinoma, and the expression level of Notch1 and VEGF is directly related to the prognosis, suggesting that the clinical detection of Notch1 and VEGF may provide a reference for assessing the severity of esophageal squamous cell carcinoma. .
