目的:检测食管癌高发区河南省林州市食管鳞癌及相应癌旁正常上皮组织的脆性组氨酸三联体(fragilehistidinetriad,FHIT)蛋白和mRNA表达情况,从而探讨FHIT与该地区食管癌发生发展的关系.方法:采用卵白素-生物素-辣根过氧化物酶复合物(ABC)免疫组织化学染色法检测了42对食管癌高发区河南省林州市手术标本的癌组织和癌旁组织中FHIT蛋白表达水平.采用反转录PCR方法检测了16对食管癌和癌旁正常组织中FHIT基因mRNA表达状况.结果:FHIT蛋白在食管癌组织中缺失表达(24/42,57.1%)或弱表达(18/42,42.9%),与癌旁正常上皮组织中FHIT蛋白情况(40/42,95.2%)差异显著(P<0.05).低分化癌组织中FHIT蛋白阴性表达率为(13/15,87%),分化癌组织阴性表达率为(11/27,41%),二者有显著性差异(P<0.05).癌组织FHITmRNA表达阳性率(4/16,25%)显著低于癌旁组织(12/16,75%)(P<0.05).结论:FHIT基因可能在该地区食管癌的发生发展过程中起重要作用,是重要的抑癌蛋白,其表达减弱或缺失增加了食管癌的易感性.
Objective: To detect the expression of fragile histidine triad (FHIT) protein and mRNA in esophageal squamous cell carcinoma of esophageal cancer and its corresponding adjacent normal epithelium in high incidence area of esophageal cancer in Henan Province, so as to explore the relationship between FHIT and the development of esophageal cancer .Methods: 42 avian-biotin-horseradish peroxidase complex (ABC) immunohistochemical staining methods were used to detect the expression of 42 cancer tissues and paracancer tissues in the surgical specimens of Linzhou, a high incidence area of esophageal cancer FHIT protein expression was detected in 16 pairs of esophageal and para-cancerous tissues by reverse transcription PCR.Results: FHIT protein was undetectable in esophageal cancer (24 / 42,57.1%) or The expression of FHIT protein in poorly differentiated carcinoma was significantly lower than that in normal adjacent epithelium (40/42, 52.2%) (P <0.05) / 15,87%), the positive rate of FHITmRNA expression was (11/27, 41%) in differentiated carcinoma (P <0.05) (12/16, 75%) (P <0.05) .Conclusion: FHIT gene may have been associated with the development of esophageal cancer in the area Play an important role, it is an important tumor suppressor protein, which lack expression increases susceptibility to esophageal cancer.
