本文介绍一种柯萨奇B_5病毒感染的搏动乳鼠心肌细胞微量模型。以400TCID_(50)/0.05ml病毒感染心肌细胞,48h后出现明显细胞病变,心肌释放酶(LDH及SGOT)明显升高,DNA合成速率明显下降;96h后,受染细胞培养液中产生Lg6.95TCID_(50)的病毒,细胞质出现特异性荧光着色。该模型证实,柯萨奇B_5病毒可在鼠心肌细胞中复制、装配并释放;同时也为诊治病毒性心肌炎的各种研究提供了工具。
This article describes a Coxsackie B_5 virus-infected rat cardiomyocyte micromodel. Cardiomyocytes were infected with 400 TCID_(50)/0.05 ml virus. After 48 h, there were obvious cytopathic changes. Myocardial release enzymes (LDH and SGOT) increased significantly, and the DNA synthesis rate decreased significantly. After 96 hours, Lg6 was produced in the cultured cells. In the 95TCID_(50) virus, the cytoplasm showed specific fluorescent staining. This model confirms that Coxsackie B_5 virus can be replicated, assembled, and released in murine cardiomyocytes; it also provides a tool for various studies of the diagnosis and treatment of viral myocarditis.
