接骨草黄酮类物质超声提取工艺优化及其抗氧化活性

目的:探讨接骨草黄酮的提取工艺及其抗氧化特性。方法:采用超声乙醇浸提法,利用单因素、正交试验对黄酮提取工艺进行优化,用水杨酸和邻二氮菲系统测定粗黄酮对羟自由基清除作用。结果:发现接骨草黄酮用55%乙醇,料液比为1∶70条件下超声时间40 min提取效果最好,最大得率为8.352 mg/g。接骨草黄酮对Fenton体系产生的·OH自由基有很好清除作用,在水杨酸体系中接骨草黄酮对羟自由基的清除效果优于芦丁,与Vc的效果相当,其抗氧化能力分别相当于Vc和芦丁的0.9987倍和5.847倍。在邻二氮菲体系中接骨草黄酮对羟自由基的清除效果优于芦丁弱于Vc,其抗氧化能力分别相当于Vc和芦丁的0.6769倍和1.169倍。 Objective: To investigate the extraction process of flavonoids from buckwheat and its antioxidative properties. Methods: Ultrasonic ethanol extraction was used to optimize the extraction process of flavonoids by single factor and orthogonal test. The scavenging effect of crude flavone on hydroxyl radical was determined by salicylic acid and phenanthroline system. Results: It was found that the extraction of buckthorn flavone with 55% ethanol, the ratio of solid to liquid 1:70 ultrasound extraction time of 40 min the best, the maximum yield was 8.352 mg / g. Scutellaria flavonoids scavenged · OH free radicals produced by Fenton system. Sclerotin was superior to rutin in the salicylic acid scavenging effect on hydroxyl free radical, which was comparable to Vc, and its antioxidant capacity Equivalent to 0.9987 times Vc and rutin 5.847 times. In ortho-phenanthroline system, the scavenging effect of glufomangone on hydroxyl radical is better than that of rutin on Vc, its antioxidant capacity is 0.6769 times and 1.169 times higher than Vc and rutin, respectively.
