目的建立消渴灵片中盐酸小檗碱含量的测定方法。方法采用高效液相色谱法,以Century SIL C18柱(250 mm×4.6mm,5μm)为色谱柱,乙腈-0.05 mol·L-1磷酸二氢钾溶液(51∶49,每1 000 ml流动相含十二烷基磺酸钠1.7 g)为流动相,流速为1 ml·min-1,检测波长为345 nm,柱温30℃,以外标法检测。结果供试品溶液色谱中,盐酸小檗碱在0.038~0.304μg范围内与峰面积呈良好线性关系,回归方程为A=4 281 910 C+3 319(r=0.999 9),平均回收率为98.01%,RSD=1.16%(n=6)。结论该方法样品分离度佳、灵敏度高、重现性好,结果准确可靠。
Objective To establish a method for the determination of berberine hydrochloride in Xiaokeling tablets. Methods High performance liquid chromatography (HPLC) was used on a Century SIL C18 column (250 mm × 4.6 mm, 5 μm) as a chromatographic column, acetonitrile-0.05 mol·L -1 potassium dihydrogen phosphate solution (51:49, Containing 1.7 g of sodium dodecyl sulfate) as the mobile phase at a flow rate of 1 ml · min-1 with a detection wavelength of 345 nm and a column temperature of 30 ° C. Results The results showed that berberine hydrochloride showed a good linear relationship with the peak area in the range of 0.038-0.304 μg, and the regression equation was A = 4 281 910 C + 3 319 (r = 0.999 9). The average recovery was 98.01%, RSD = 1.16% (n = 6). Conclusion The method has the advantages of good resolution, high sensitivity, good reproducibility and accurate and reliable results.
