目的探讨天然冰片及其结构修饰物对戊四唑所致惊厥模型小鼠氨基酸类神经递质含量的影响。方法采用戊四唑制备小鼠惊厥模型,观察给药后动物惊厥的发生及其潜伏时间,再通过高效液相色谱-荧光检测法测定脑内氨基酸类神经递质的含量。结果与模型组比较,天然冰片高剂量组小鼠抗惊厥数量显著增加,惊厥潜伏期显著延长;(+)-顺丁烯二酸单冰片酯中剂量组惊厥潜伏期显著延长。与模型组比较,天然冰片低剂量组脑内天冬氨酸(ASP)显著增加、谷氨酸(Glu)含量显著减少;天然冰片高剂量组γ-氨基丁酸(GABA)含量显著增加,(+)-顺丁烯二酸单冰片酯中剂量组Glu含量显著减少。结论天然冰片及其结构修饰物可能通过改变脑内氨基酸类神经递质的含量来改善模型小鼠的惊厥行为。
Objective To investigate the effects of natural borneol and its structural modifications on amino acid neurotransmitters in pentylenetetrazole-induced convulsions model mice. Methods The pentylenetetrazole was used to prepare the convulsions model in mice. The occurrence and latency of convulsions in the animals were observed. The contents of amino acid neurotransmitters in the brain were determined by high performance liquid chromatography-fluorescence detection. Results Compared with the model group, the amount of anticonvulsant increased significantly and the latent period of convulsion increased significantly in the high dose of natural borneol high dose group. The latent period of convulsion of (+) - monobasic maleate medium dose group was significantly prolonged. Compared with the model group, the content of aspartate (ASP) and glutamic acid (Glu) in the low-dose natural borneol group increased significantly; the content of GABA in the high-dose natural borneol group increased significantly +) - maleic acid monobutyric acid ester dose group Glu content was significantly reduced. Conclusion Natural borneol and its structural modifications may improve the convulsant behavior of model mice by changing the content of amino acid neurotransmitters in the brain.
