目的研究北京地区儿童ORMDL3基因表达水平、生活方式、室内空气质量与哮喘发生的相关性。方法采用病例-对照方法进行实验设计。使用RT-PCR法检测儿童ORMDL3基因表达水平;通过《国际儿童哮喘及过敏研究(ISSAC)问卷》收集儿童生活方式信息;并根据《室内空气质量标准(GB/T18883-2002)》中规定的检验方法对室内空气中PM2.5,VOCs,甲醛浓度及空气温湿度进行监测。使用t检验、卡方检验分析病例组与对照组儿童ORMDL3基因表达水平,生活方式与室内空气质量差异;使用Logistic逐步回归来分析ORMDL3基因表达水平,生活方式与室内空气质量对与哮喘之间相关性。结果 ORMDL3基因表达水平在病例组与对照组中有显著差异(P<0.05)。在早发型哮喘患儿中,ORMDL3基因表达水平、哮喘家族史与哮喘显著相关(P<0.05)。而在迟发型哮喘患儿中,ORMDL3基因表达水平、采暖方式、室内空气湿度与哮喘显著相关(P<0.05)。结论 ORMDL3基因表达水平与儿童哮喘的发生密切相关,早发型哮喘患儿与遗传因素有显著关联,而迟发型哮喘患儿中遗传因素与环境因素均与哮喘相关。
Objective To investigate the correlation between ORMDL3 gene expression, lifestyle, indoor air quality and asthma in children in Beijing. Methods Case-control method was used to design the experiment. The level of ORMDL3 gene expression in children was detected by RT-PCR. The lifestyle information of children was collected through the International Children’s Asthma and Allergy Study (ISSAC) questionnaire. According to the test specified in Indoor Air Quality Standard (GB / T 18883-2002) Methods The indoor air PM2.5, VOCs, formaldehyde concentration and air temperature and humidity were monitored. Logistic regression was used to analyze ORMDL3 gene expression levels, lifestyle and indoor air quality related to asthma, using t-test and chi-square test to analyze ORMDL3 gene expression level, lifestyle and indoor air quality in case group and control group Sex. Results ORMDL3 gene expression levels in the case group and control group were significantly different (P <0.05). In children with early-onset asthma, ORMDL3 gene expression, family history of asthma and asthma were significantly correlated (P <0.05). In children with delayed type asthma, ORMDL3 gene expression, heating mode, indoor air humidity and asthma were significantly correlated (P <0.05). Conclusions ORMDL3 gene expression is closely related to the development of asthma in children. Early onset asthma children are significantly associated with genetic factors. However, both genetic and environmental factors in children with delayed asthma are associated with asthma.
