目的 探讨血管内皮生长因子(VEGF) 及其受体(KDR) 的表达与人胃癌组织血管生成的关系。方法 采用免疫组织化学ABC法观察了50 例人胃癌组织中的VEGF及KDR 的定位与分布,并对血管进行染色及计数。结果 50 % 的胃癌组织表达VEGF,阳性物质主要位于肿瘤细胞胞膜及胞浆,KDR 主要位于癌组织及癌组织旁的血管内皮细胞。VEGF 表达阳性胃癌组织的血管密度(13-91 ±7-24) 显著高于VEGF表达阴性者(6-50 ±2-10)( P< 0-05) ,而且随着VEGF表达强度的增强,癌组织内血管密度明显增加。结论 胃癌细胞分泌的VEGF主要以旁分泌的形式促进肿瘤血管的生成,也可能存在着自分泌形式。
Objective To investigate the relationship between the expression of vascular endothelial growth factor (VEGF) and its receptor (KDR) and angiogenesis in human gastric cancer. Methods Immunohistochemical ABC method was used to observe the localization and distribution of VEGF and KDR in 50 human gastric cancer tissues, and to stain and count blood vessels. Results 50% of gastric cancer tissues expressed VEGF, positive substances mainly located in the membrane and cytoplasm of tumor cells, and KDR mainly located in vascular endothelial cells adjacent to cancer tissues and cancer tissues. Vascular density (13-91 ±7-24) in VEGF-positive gastric cancer tissues was significantly higher than that in VEGF-negative humans (6-50 ±2-10) (P < 0-05), and with the increase of VEGF expression intensity, The density of blood vessels in the cancer tissue increased significantly. Conclusion VEGF secreted by gastric cancer cells mainly promotes the formation of tumor vasculature in the form of paracrine, and there may also be autocrine forms.
