观察了3种细胞因子和顺式维甲酸(c-RA)在体外对U_(937)细胞增殖与分化的影响。结果表明:rhTNF-α、PHA-LCM和c-RA均能抑制U_(937)细胞的增殖并诱导其向成熟细胞分化,作用方式呈剂量和时间依赖性,而且rhTNF-α和c-RA合用后作用增强。rhIL-1-α单独对U_(937)细胞的增殖与分化没有影响,但可增强c-RA的诱导分化作用。[~3H]-TdR掺入实验表明,rhTNF-α、PHA-LCM和c-RA通过抑制U_(937)细胞合成DNA而使其增殖停滞。结果揭示:合理联合应用分化诱导剂能互相增强作用,对进一步提高白血病细胞的诱导分化效应以及临床治疗有一定的参考价值。
The effects of three cytokines and cis-retinoid (c-RA) on the proliferation and differentiation of U_(937) cells in vitro were observed. The results showed that rhTNF-α, PHA-LCM and c-RA all inhibited the proliferation of U_(937) cells and induced their differentiation into mature cells. The mode of action was dose- and time-dependent, and rhTNF-α and c-RA were combined. After the role of enhanced. RhIL-1-α alone had no effect on the proliferation and differentiation of U_(937) cells, but it could enhance the induction and differentiation of c-RA. [~3H]-TdR incorporation experiments showed that rhTNF-α, PHA-LCM and c-RA inhibited the proliferation of U_(937) cells through the synthesis of DNA. The results revealed that: The rational use of differentiation inducer can enhance each other’s effect, and it has certain reference value for further improving the induction and differentiation of leukemia cells and clinical treatment.
