目的观察蛋白前体转化酶Furin在慢性宫颈炎、各级宫颈上皮内瘤变(CIN)及宫颈鳞状细胞癌(CSCC)中的表达差别,研究其作为宫颈癌早期筛查的临床意义。方法按照常规方法提取研究对象180例宫颈各类病变组织中的蛋白,通过Western Blot检测Furin蛋白表达情况,并行人乳头状瘤病毒分型检测。结果 180例标本中,Furin蛋白阳性表达率为65.6%,其中低级别子宫颈上皮内瘤变(CIN-L)组与慢性宫颈炎组比较差异有统计学意义(P<0.05);低级别宫颈上皮内瘤变(CIN-L)组与CIN-H组比较差异有统计学意义(P<0.05);高级别CIN-H组与CSCC组比较差异无统计学意义(P>0.05)。180例人乳头状瘤病毒(HPV)组中,高危型HPV检出率为76.1%,低危型为12.2%。高危型HPV在慢性宫颈炎组、CIN组、CSCC组检出率逐渐升高,差异有统计学意义(P<0.05),其中CIN-L组与CIN-H组比较差异无统计学意义(P>0.05)、CIN-H组与CSCC组比较差异不显著(P>0.05)。结论随着宫颈病变级别的升高Furin蛋白的表达呈增加趋势,有助于预测宫颈病变的进展情况,高危型HPV感染与Furin蛋白阳性表达呈正相关(P<0.05),二者联合检测是宫颈癌早期筛查的理想手段。
Objective To observe the expression of furin in chronic cervicitis, cervical intraepithelial neoplasia (CIN) and cervical squamous cell carcinoma (CSCC) of chronic cervicitis, and to study its clinical significance as an early screening of cervical cancer. Methods According to the routine method, 180 cases of cervical lesions were extracted from the diseased tissues. The expression of Furin protein was detected by Western Blot and the human papillomavirus type was detected. Results The positive rate of Furin protein expression was 65.6% in 180 cases of specimens, of which the difference between CIN-L group and chronic cervicitis group was statistically significant (P <0.05). The low grade cervix The difference between CIN-H group and CIN-H group was statistically significant (P <0.05). There was no significant difference between CIN-H group and CSCC group (P> 0.05). Among 180 cases of human papilloma virus (HPV), the detection rate of high-risk HPV was 76.1% and low-risk type was 12.2%. The detection rate of high-risk HPV in chronic cervicitis group, CIN group and CSCC group was significantly higher than that in CIN-H group (P <0.05), and there was no significant difference between CIN-L group and CIN-H group > 0.05). There was no significant difference between CIN-H group and CSCC group (P> 0.05). Conclusion The expression of Furin increases with the increase of cervical lesions, which is helpful to predict the progression of cervical lesions. The positive rate of high-risk HPV infection is positively correlated with Furin protein (P <0.05) The ideal means of early cancer screening.
