目的:建立测定卷柏类药材中双黄酮成分的高效液相色谱方法。方法:色谱柱:Elite-ODS(5 μm,150 mm×4.6mm);流动相:甲醇-0.050 mol·L~(-1)磷酸二氢钠溶液系统,梯度洗脱,0→6 min,甲醇52.5%→70.0%;柱温:40℃;流速:1.5 mL·min~(-1),检测波长:330 nm。结果:穗花杉双黄酮、扁柏双黄酮、银杏双黄酮加样回收率分别为97.05%,98.45%,96 88%(RSD=2.1%,2.8%,2.3%,n=3)。卷柏类药材均含有穗花杉双黄酮,但含量差异较大;另2种双黄酮成分在卷柏类药材中的分布与含量同样存在差异。结论:本法快捷、灵敏、准确,可作为评价卷柏类药材的参考依据。
Objective: To establish a HPLC method for the determination of biflavonoids in Selaginella. Method: Column: Elite-ODS (5 μm, 150 mm×4.6 mm); mobile phase: methanol-0.050 mol·L-1 sodium dihydrogen phosphate solution, gradient elution, 0→6 min, methanol 52.5%→70.0%; column temperature: 40°C; flow rate: 1.5 mL·min -1; detection wavelength: 330 nm. Results: The recovery rates of spiked flavonoids, cypress flavonoids and ginkgo biflavonoids were 97.05%, 98.45% and 96 88%, respectively (RSD=2.1%, 2.8%, 2.3%, n=3). The Selaginella species contained both flavonoids, but the content was different. The distribution and content of the other two flavonoids in Selaginella were also different. Conclusion: This method is rapid, sensitive and accurate and can be used as a reference for evaluating medicinal materials of Selaginella.
