采用人工授精法以河川沙塘鳢(Odontobutis potamophila)为母本、鸭绿沙塘鳢(O.yaluensis)为父本进行杂交,获得正常发育的杂交子代。利用胸腔活体注射植物凝集素(PHA)和秋水仙素溶液后,取头肾细胞进行低渗处理,空气干燥法制片,对河川沙塘鳢、鸭绿沙塘鳢及其杂交子代的核型进行了比较研究。结果表明:河川沙塘鳢的核型为2n=44,2st+42t,NF=44;鸭绿沙塘鳢的核型为2n=44,2st+42t,NF=44;杂交子代的核型为2n=44,4st+40t,NF=44。经对比分析,杂交子代分别继承了双亲的一套染色体,表明杂交子代为真正的杂交种。
Using artificial insemination method, we used the Odontobutis potamophila as the female parent and O. yaruensis as the male parent to obtain the hybrid progeny of normal development. After injecting phytohemagglutinin (PHA) and colchicine solution into the thoracic cavity, the kidney cells were harvested for hypotonic treatment and air-dried method. The karyotypes of S. sativa, A. japonicus and their hybrid progeny Conducted a comparative study. The results showed that the karyotypes of 2 species were 2n = 44st + 42t, NF = 44, and 2n = 44st + 42t, NF = 44. The karyotype of hybrids 2n = 44,4st + 40t, NF = 44. After comparative analysis, the hybrid progeny inherited a set of chromosomes of the parents respectively, indicating that the hybrid progenies are the true hybrids.
