目的:探讨甘氨双唑钠和塞来昔布联合同步放化疗(顺铂10mg/L+放疗2Gy)对宫颈癌Siha细胞增殖的影响。方法:用不同浓度的甘氨双唑钠及塞来昔布分别处理Siha细胞24h后,MTT法检测细胞的增殖活性,并求出无毒浓度;用两药的无毒浓度分别及同时联合同步放化疗处理Siha细胞24h后,MTT法检测细胞的增殖活性,倒置显微镜及透射电镜观察细胞形态的改变。结果:联合组对细胞增殖的抑制作用比同步放化疗组更显著(P<0.01),且两药联合组的抑制作用强于单药联合组及同步放化疗组(P<0.01);倒置显微镜及透射电镜下两药联合组细胞呈现出凋亡形态。结论:甘氨双唑钠和塞来昔布对Siha细胞同步放化疗有增敏作用,且两药与同步放化疗联合应用有一定的协同效应。
Objective: To investigate the effect of glycopyrrolate sodium and celecoxib combined with chemoradiotherapy (cisplatin 10mg / L + radiotherapy 2Gy) on the proliferation of cervical cancer Siha cells. Methods: Siha cells were treated with different concentrations of glycopyrrolate sodium and celecoxib for 24 h respectively. MTT assay was used to detect the proliferative activity of the cells and the non-toxic concentration was determined. The chemotherapeutic activity of Siha cells was determined by MTT assay after 24h. The morphological changes of cells were observed by inverted microscope and transmission electron microscope. Results: The inhibitory effect of the combination group was more significant than that of the concurrent chemoradiotherapy group (P <0.01), and the inhibitory effect of the combination group was stronger than that of the single drug combination group and concurrent chemoradiotherapy group (P <0.01) And the transmission electron microscope showed that the combination of two drugs showed apoptotic morphology. CONCLUSION: Glycine diazoxide sodium and celecoxib have sensitizing effects on Siha synchrotron chemoradiation and the synergistic effect of the two drugs in combination with concurrent chemoradiotherapy.
