目的探讨膜联蛋白(annexin)Ⅳ在月经周期子宫内膜组织的表达规律。方法选择2006年10月至2007年8月在中山大学附属第一医院妇产科生殖医学中心因男方原因要求再生育的18名女性志愿者,采用免疫组化链菌生物素蛋白-过氧化物酶连接法(SP法)和Western blot方法对月经周期子宫内膜组织进行膜联蛋白Ⅳ定位与半定量表达的实验研究。结果免疫组化实验显示增殖期和分泌早期子宫内膜上皮细胞均存在膜联蛋白Ⅳ蛋白表达,分泌中、晚期上皮和基质细胞中均有表达。分泌中、晚期的子宫内膜组织膜联蛋白Ⅳ的表达量较增殖中、晚期显著增高(膜联蛋白Ⅳ与β-actin条带平均灰度值比值结果分别为0.324、0.369对0.231、0.127),P<0.05;分泌早期膜联蛋白Ⅳ表达量(0.152)无明显增高;膜联蛋白Ⅳ表达量与LH峰值无显著相关性,分泌期子宫内膜膜联蛋白Ⅳ表达量与孕激素水平相关。结论膜联蛋白Ⅳ可能是参与月经周期子宫内膜形态与功能转化的重要蛋白质。
Objective To investigate the expression of annexin IV in endometrial tissue during menstrual cycle. METHODS: From October 2006 to August 2007, 18 female volunteers who were required to be reborn for male reasons at the Obstetrics and Gynecology Reproductive Medicine Center of the First Affiliated Hospital of Sun Yat-sen University were enrolled in this study. Immunohistochemical streptavidin-peroxide Enzyme-linked immunosorbent assay (SP method) and Western blot method were used to study the location and semiquantitative expression of Annexin IV in endometrial tissues of menstrual cycle. Results Immunohistochemistry showed that Annexin IV protein was expressed in proliferative and early secretory endometrial epithelial cells, both in secretory and epithelial cells and in stromal cells. Secretion of late and endometrial annexin IV expression levels were significantly higher than in the late proliferative (Annexin Ⅳ and β-actin bands with the average gray value of the results were 0.324,0.369 pairs 0.231,0.127) , P <0.05; early secretion of annexin Ⅳ expression (0.152) was not significantly increased; annexin Ⅳ expression level and LH peak no significant correlation between secretory endometrial annexin Ⅳ expression and progesterone levels . Conclusions Annexin IV may be involved in menstrual cycle endometrial morphology and function of the important proteins.
