血清缺乏是在细胞治疗中对供体BMSCs活性影响的重要因素之一。作者探讨了不同浓度血清对MSCs细胞凋亡的影响。体外培养大鼠MSCs,用不同浓度血清(FBS)处理MSCs,MTT法检测MSCs细胞生长曲线,用Hoechst33342/PI染色检查MSCs的细胞凋亡,并使用Western blot法分析细胞凋亡相关生物信号分子的表达。结果表明,血清缺乏培养条件下,MSCs在培养72h内出现细胞凋亡。并且,无血清培养直接引起了Casepase-3,Casepase-8和Casepase-9蛋白质表达量的增加,导致细胞凋亡的发生,从而致使细胞生长曲线的下降。此外,高浓度的血清对细胞凋亡具有保护作用。
Serum deficiency is one of the important factors that influence donor BMSCs activity in cell therapy. The authors explored the effects of different concentrations of serum on apoptosis of MSCs. The MSCs were cultured in vitro and MSCs were treated with different concentrations of serum (FBS). The growth curve of MSCs was detected by MTT assay. The apoptosis of MSCs was examined by Hoechst33342 / PI staining. Western blot was used to analyze the expression of apoptosis related biomarkers expression. The results showed that under the condition of serum-free culture, MSCs appeared apoptosis within 72h. In addition, serum-free culture directly induced an increase in the expression of Casepase-3, Casepase-8 and Casepase-9 proteins, leading to apoptosis, resulting in a decrease in cell growth curve. In addition, high concentrations of serum have a protective effect on apoptosis.
