目的:利用蛋白质组学方法建立膀胱低级别尿路上皮癌组织及其癌旁黏膜组织的差异蛋白质表达谱。方法:对15例人低级别尿路上皮癌组织和配对的癌旁黏膜组织进行比较蛋白质组学研究,利用双向凝胶电泳(2-DE)分离两者总蛋白质后,经图像分析识别差异表达的蛋白,应用基质辅助激光解吸电离飞行时间质谱(MALDI-TOF-MS)鉴定差异蛋白质。结果:1)比较分析15例膀胱癌及正常配对组织的2-DE图谱,得到差异蛋白质点54个;2)对54个差异蛋白质点进行了肽质量指纹图分析,鉴定出一些与癌基因、细胞周期调控及信号转导等有关的膀胱癌相关蛋白。结论:成功鉴定45个膀胱低级别尿路上皮癌组织相关蛋白,为进一步筛选用于膀胱癌诊断、治疗和预后评估的分子标志奠定基础。
OBJECTIVE: To establish differential protein expression profiles of low-grade bladder urothelial carcinoma and its adjacent mucosa using proteomics. Methods: Fifteen cases of low-grade urothelial carcinoma and matched paracancerous mucosa tissues were studied by comparative proteomics. The two proteins were separated by two-dimensional gel electrophoresis (2-DE), and then differentially expressed by image analysis The proteins were identified by matrix-assisted laser desorption / ionization time-of-flight mass spectrometry (MALDI-TOF-MS). Results: 1) Comparative analysis of 2-DE maps of 15 cases of bladder cancer and normal matched tissues, 54 differential proteins were obtained; 2) 54 different protein spots were analyzed by peptide mass fingerprinting to identify some of the cancer genes, Cell cycle regulation and signal transduction and other related bladder cancer related proteins. CONCLUSION: The successful identification of 45 low-grade bladder urothelial carcinoma tissue-related proteins will lay the foundation for further screening molecular markers for diagnosis, treatment and prognosis of bladder cancer.
