基于黄瓜果实转录组学研究,筛选并克隆了一个扩张素蛋白(β-expansin)基因Cs EXPb1。蛋白序列同源比对发现Cs EXPb1蛋白是一类钙调素类似蛋白(Calmodulin-like Protein,CML),与甜瓜、草莓及番茄扩张素蛋白基因具有较高的相似性;Ex Pasy分析发现Cs EXPb1为亲水性蛋白,亚细胞定位分析证明该蛋白广泛分布在细胞质以及细胞核结构中;q RT-PCR分析发现Cs EXPb1无论是在授粉坐果还是单性结实坐果过程中皆上调表达,而在不授粉败育的果实中下调表达;q RT-PCR也表明Cs EXPb1的表达受到生长素、细胞分裂素和赤霉素等单性结实诱导激素的正向调控,推测Cs EXPb1与黄瓜坐果有关,可能参与了激素诱导的单性结实过程。
Based on the study of cucumber fruit transcriptomics, Cs-EXPb1, a β-expansin gene, was screened and cloned. Protein sequence homology comparison revealed that Cs-EXPb1 protein is a kind of calmodulin-like protein (CML), which has high similarity with melon, strawberry and tomato expandin protein genes. Ex Pasy analysis found that Cs-EXPb1 As a hydrophilic protein, subcellular localization analysis showed that the protein was widely distributed in the cytoplasm and the nuclear structure; q RT-PCR analysis showed that Cs-EXPb1 was up-regulated both in pollination and fruit setting, Q RT-PCR also showed that the expression of Cs-EXPb1 was positively regulated by auxin, cytokinin, and gibberellin, and so on, suggesting that Cs-EXPb1 may be involved in cucumber fruit setting and may be involved in Hormone-induced parthenocarpic processes.
