利用常规杂交瘤技术制备了2株抗人绒毛膜促性腺激素(hCG)的杂交瘤细胞。ELISA结果表明,单克隆抗体ID5和4D5均与人hCG有特异性反应,腹水效价分别为3.2×10-5、6.4×10-5,均属于IgG1亚类。与人促卵泡激素(hFSH)和人促黄体激素(hLH)的交叉反应呈阴性。免疫转印结果显示能识别相对分子质量为24×103u的hCG-β链条带。亲和常数分别为1.01×107L/mol、4.43×107L/mol。
Two anti-human chorionic gonadotropin (hCG) hybridoma cells were prepared by conventional hybridoma technique. The results of ELISA showed that the monoclonal antibodies ID5 and 4D5 both reacted specifically with human hCG. The ascites titer was 3.2 × 10-5 and 6.4 × 10-5, respectively, belonging to IgG1 subclass. Cross-reactivity with human follicle-stimulating hormone (hFSH) and human luteinizing hormone (hLH) was negative. Immunoblotting showed that the hCG-β chain with a molecular weight of 24 × 103u could be identified. The affinity constants were 1.01 × 107 L / mol and 4.43 × 107 L / mol, respectively.
