目的观察消岩汤对环磷酰胺诱导Lewis肺癌小鼠造血微环境的影响,探究消岩汤对Lewis肺癌小鼠骨髓抑制的造血微环境的改善作用及其作用机制。方法 SPF级Lewis肺癌小鼠随机分为对照组、模型组、重组人粒细胞刺激因子组和消岩汤组,每组各10只。对照组和模型组ig 0.02 mL/g生理盐水。人粒细胞刺激因子组在造模后sc重组人粒细胞刺激因子注射液15 ng/g,1次/d,连续给药7 d。消岩汤组在造模后ig 2.9 g/mL消岩汤,2次/d,0.2 mL/次,连续给药7 d。采用流式细胞仪分析细胞周期,通过细胞培养技术检测基质细胞分化能力,采用QRT-PCR法检测骨髓细胞Bcl-2和Bax mRNA的基因表达。结果与模型组比较,消岩汤能促进骨髓细胞从G_0/G_1期向S、G2/M期转化(P<0.05);能一定程度上缓解化疗导致的成纤维细胞集落生成单位(CFU-F)形成减少(P<0.05);能上调抗凋亡基因Bcl-2 mRNA的表达((P<0.05)。结论消岩汤能够促进骨髓细胞向增殖周期转化,上调Bcl-2 mRNA表达,改善造血微环境,从而缓解Lewis肺癌骨髓抑制对小鼠造血功能的影响。
Objective To observe the effect of Xiao Yan Decoction on the hematopoietic microenvironment induced by cyclophosphamide in mice with Lewis lung cancer and to explore the effect and mechanism of “Xiao Yan Decoction” on the hematopoietic microenvironment of Lewis lung carcinoma in mice. Methods SPF Lewis lung cancer mice were randomly divided into control group, model group, recombinant human granulocyte-stimulating factor group and HuanYanTang group, with 10 mice in each group. The control group and model group ig 0.02 mL / g saline. Human granulocyte-stimulating factor group was injected with 15g / g sc recombinant human granulocyte stimulating factor once a day for 7 days. Xiao Yan Decoction ig 2.9g / mL rock soup after modeling, 2 times / d, 0.2 mL / time, continuous administration of 7 d. The cell cycle was analyzed by flow cytometry, the differentiation of stromal cells was detected by cell culture technique, and the gene expression of Bcl-2 and Bax mRNA was detected by QRT-PCR. Results Compared with the model group, Xiaoyantang can promote the transformation of bone marrow cells from G_0 / G_1 phase to S, G2 / M phase (P <0.05), alleviate the fibroblast colony forming unit (CFU-F (P <0.05) .Conclusion: QIANGYAN Decoction can promote the transformation of myeloid cells to the proliferative cycle, up-regulate the expression of Bcl-2 mRNA and improve the hematopoietic Microenvironment, thereby alleviating Lewis lung cancer bone marrow suppression on hematopoietic function.
