目的 探讨MMP - 2在EAM发病中的作用 ,以及甲基强的松龙对MMP - 2基因mRNA和蛋白表达的影响。方法 应用RT -PCR及免疫组织化学技术 ,检测MMP - 2在EAM组、甲基强的松龙治疗组 (EAMM组 )和对照组大鼠外周血、脾脏、肌肉组织中的表达水平。结果 ⑴EAMM组大鼠发病程度低于EAM组 ,炎症细胞浸润和肌纤维坏死程度减轻。⑵MMP - 2在EAM大鼠外周血、脾脏淋巴细胞中的表达上调 ;MMP - 2蛋白在EAM组大鼠肌肉组织表达增加 ,与对照组比较 ,差异有显著性。EAMM组MMP - 2mRNA的表达及蛋白的表达均受到抑制 ,与EAM组比较差异有显著性。结论 MMP - 2表达增加可能与EAM发病有关 ;甲基强的松龙能减轻EAM发病程度 ,其作用机理可能与MMP - 2的表达下调有关
Objective To investigate the role of MMP - 2 in the pathogenesis of EAM and the effect of methylprednisolone on the expression of MMP - 2 mRNA and protein. Methods RT - PCR and immunohistochemistry were used to detect the expression of MMP - 2 in peripheral blood, spleen and muscle tissue of EAM group, methylprednisolone group (EAMM group) and control group. Results (1) The incidence of EAMM rats was lower than EAM group, inflammatory cell infiltration and muscle fiber necrosis reduced. (2) The expression of MMP - 2 in peripheral blood and spleen lymphocytes of EAM rats was up - regulated; the expression of MMP - 2 protein in muscle tissue of EAM rats increased compared with that in control group, the difference was significant. EAMM group MMP - 2 mRNA expression and protein expression were inhibited, compared with the EAM group was significantly different. Conclusion The increased expression of MMP - 2 may be related to the pathogenesis of EAM. Methylprednisolone may reduce the incidence of EAM, and its mechanism may be related to the down - regulation of MMP - 2.
