目的建立多发性硬化(MS)患者用药前后脑脊液蛋白质组一维、二维液相色谱分离的技术体系。方法以MS患者用药前后脑脊液为研究对象,提取脑脊液蛋白质组,分别采用超高压液相色谱-四级杆-飞行时间质谱(UPLC-Q-TOFMS)技术和二维色谱聚焦-反向液相色谱(PF-2D)技术进行分离鉴定。结果分别获得MS患者用药前后脑脊液蛋白质组一维和二维液相色谱图谱,分离方法有良好的重现性。筛选并鉴定出表达有明显差异的蛋白质。结论用液相色谱技术筛选差异蛋白有助于寻找具有临床治疗意义的MS疾病标识物,为MS疾病的早期诊断和治疗提供理论和实验依据。
Objective To establish a one-dimensional and two-dimensional liquid chromatographic separation system for cerebrospinal fluid proteome in patients with multiple sclerosis (MS) before and after treatment. Methods The cerebrospinal fluid (CSF) was collected from patients with MS before and after the treatment. The proteome of cerebrospinal fluid (CSF) was extracted and purified by UPLC-Q-TOFMS and 2D-Focused Reverse-phase Liquid Chromatography (PF-2D) technology for isolation and identification. Results One-dimensional and two-dimensional liquid chromatograms of cerebrospinal fluid proteome were obtained before and after treatment of MS patients respectively. The separation method showed good reproducibility. Screen and identify the expression of significant differences in the protein. Conclusion The screening of differential proteins by liquid chromatography is helpful to find MS markers with clinical significance and provide theoretical and experimental evidences for the early diagnosis and treatment of MS diseases.
