目的探讨IgG在喉鳞癌细胞中的表达及其与B淋巴细胞源性IgG的差异性。方法采用免疫组化、原位杂交法检测IgG在喉鳞癌细胞的表达;激光显微切割结合RT-PCR法检测喉鳞癌细胞表达IgG重链、轻链基因;测序并BLAST比对IgG重链可变区的VDJ重排;分析喉鳞癌表达IgG与临床病理参数的关系。结果重链Igγ、轻链Igκ和Igλ蛋白在同一癌细胞中都有表达,且主要分布于癌巢的外围;IgG重链恒定区IGHG1基因m RNA的表达与Igγ在癌细胞中共定位;喉鳞癌细胞中IGHG1、Igκ及Igλ基因均有表达;癌细胞中IgG的VDJ重排呈特定的模式,且有少量的突变,突变率为2.03%~3.67%;低分化、Ⅲ-Ⅳ期或有淋巴结转移的病例有更强的IgG表达。结论喉鳞癌细胞能够表达IgG,其不同于B淋巴细胞源性IgG的VDJ重排机制;喉鳞癌细胞表达的IgG与临床分期、病理分级和淋巴结转移有关。
Objective To investigate the expression of IgG in laryngeal squamous cell carcinoma (LSCC) and its difference with B lymphocyte-derived IgG. Methods Immunohistochemistry and in situ hybridization were used to detect the expression of IgG in laryngeal squamous cell carcinoma. Laser microdissection and RT-PCR were used to detect the expression of IgG heavy chain and light chain genes in laryngeal squamous cell carcinoma. VDJ rearrangement of the variable region of the chain. The relationship between IgG expression in laryngeal squamous cell carcinoma and clinicopathological parameters was analyzed. Results The heavy chain Igγ, light chain Igκ and Igλ protein were expressed in the same cancer cells, and mainly distributed in the periphery of the cancer nest. The expression of m RNA of IGHG1 gene in IgG heavy chain constant region was colocalized with Igγ in cancer cells. The expression of IGHG1, Igκ and Igλ genes were all detected in cancer cells. There was a specific pattern of IgG rearrangement in cancer cells with a small number of mutations with a mutation rate of 2.03% -3.67%. The poorly differentiated, Ⅲ-Ⅳ or Cases of lymph node metastases have stronger IgG expression. Conclusions Laryngeal squamous carcinoma cells can express IgG, which is different from the VDJ rearrangement mechanism of B lymphocyte-derived IgG. The IgG expressed in laryngeal squamous cell carcinoma is related to clinical stage, pathological grade and lymph node metastasis.
