目的:探讨榄香烯乳对大鼠胃粘膜癌变发生发展中c-myc基因表达及端粒酶活性的影响。方法:采用MNNG、10%NaCl混合液加酒精灌胃建立大鼠胃癌模型,设立正常组、模型组及榄香烯组,榄香烯组在造模过程中采用榄香烯乳干预。30周后,分别采用原位杂交、免疫组化及端粒酶微孔板杂交法检测大鼠胃粘膜c-myc基因mRNA、蛋白表达及端粒酶活性。结果:榄香烯组癌变发生率低于模型组。c-myc基因表达与端粒酶活性随着胃粘膜癌变的发展呈递增趋势,并且二者具有高度相关性。结论:下调c-myc基因表达,抑制端粒酶活性,是榄香烯乳重要的抗癌机制之一。
Objective: To investigate the effect of elemene on c-myc gene expression and telomerase activity in gastric carcinogenesis. Methods: The model of gastric cancer was established by intragastric administration of MNNG and 10% NaCl and alcohol. The normal group, model group and elemene group were established. The elemene group was treated with elemene during the modeling process. After 30 weeks, the mRNA and protein expression of c-myc and the activity of telomerase in rat gastric mucosa were detected by in situ hybridization, immunohistochemistry and telomerase microplate hybridization. Results: The incidence of carcinogenesis in elemene group was lower than that in model group. The expression of c-myc and telomerase activity showed an increasing trend with the development of gastric mucosal carcinogenesis, and the two were highly correlated. Conclusion: Down-regulation of c-myc gene expression and inhibition of telomerase activity is one of the important anticancer mechanisms of elemene.
