目的:观察桃红四物汤对早期骨筋膜室综合征模型家兔血清肌酸激酶(CK)、乳酸脱氢酶(LDH)、谷草转氨酶(AST)、丙二醛(MDA)浓度和超氧化物歧化酶(SOD)含量的影响,为桃红四物汤的临床应用提供实验依据。方法:本实验选用30只健康日本大耳成年白兔,随机分为3组,10只为空白对照组,余20只参照改良“Aydin F止血带法”复制兔小腿骨筋膜室综合征模型,造模成功后随机分为桃红四物汤组(10只)和模型组(10只),桃红四物汤组予桃红四物汤灌胃(2.8 g/kg),模型组予等剂量生理盐水灌胃。分别于实验前、实验后2 h、24 h、48 h、6 d、1周共6个时间点连续动态采兔耳缘静脉血,检测CK、LDH、AST、SOD和MDA活性;实验结束后立即处死所有动物,取健侧和伤侧小腿肌肉进行病理切片检查。结果:造模后与造模前比较,兔伤侧小腿肿胀程度明显,实验后2 h,桃红四物汤组和模型组血中CK、LDH、AST和MDA水平均高于空白对照组,SOD低于空白对照组,差异均有统计学意义(P<0.01);实验后24 h至1周桃红四物汤组血中CK、LDH、AST和MDA水平各时间点均低于模型组,SOD水平各时间点均高于模型组,差异均有统计学意义(P<0.01);光镜检查结果显示:桃红四物汤组组织结构改变较模型组轻。结论:桃红四物汤能上调早期骨筋膜室综合征模型家兔血清SOD含量,抑制氧自由基的过度生成,降低血中CK、LDH、AST含量,从而改善细胞损伤。
Objective: To observe the effect of Taohong Siwu decoction on serum creatine kinase (CK), lactate dehydrogenase (LDH), aspartate aminotransferase (AST), malondialdehyde (MDA) and superoxide The content of dismutase (SOD), to provide experimental evidence for the clinical application of Taohong Siwu Decoction. Methods: Thirty healthy Japanese white rabbits were randomly divided into three groups, 10 were blank control group, the other 20 were referred to the modified “Aydin F tourniquet method” The models were randomly divided into two groups: Taohong Siwu Decoction (n = 10) and model group (n = 10). Taohong Siwu Decoction was administrated with Taohong Siwu Decoction (2.8 g / kg) Dose normal saline gavage. LDH, AST, SOD and MDA were measured continuously at 6 time points before experiment, 2 h, 24 h, 48 h, 6 d and 1 week after the experiment. At the end of the experiment All animals were immediately sacrificed, and the contralateral and injured calf muscles were examined for pathology. Results: Compared with that before the model making, the degree of swelling of the calf on the injury side of the rabbit was obvious. The levels of CK, LDH, AST and MDA in the Taohong Siwu Decoction group and the model group were significantly higher than those in the blank control group 2 h after the experiment (P <0.01). The levels of CK, LDH, AST and MDA in blood of Taohongsiwu Decoction group were lower than those of the model group at 24 h to 1 week after the experiment (P <0.01). The results of light microscopy showed that the changes of tissue structure in Taohong Siwu Decoction group were lighter than those in model group. CONCLUSION: Taohongsiwutang can upregulate the content of SOD in rabbits with early compartment syndrome, inhibit the overproduction of oxygen free radicals, and decrease the content of CK, LDH and AST in blood, so as to improve cell injury.
