应用自组装膜技术在压电石英晶振金电极表面自组装一带羧基的巯基丙酸单层膜 ,通过盐酸 1 -乙基 -3 -(3 -二甲基氨基丙基 )碳二亚胺及 N -羟基琥珀酰亚胺共价固定 3 2 KD的日本血吸虫分子抗原 (Sj Ag3 2 ) ,设计了石英晶振微天平免疫传感器 ,用于测定日本血吸虫抗体 .比较了巯基自组装单层膜与 HEMA-MMA共聚物涂层修饰的石英晶振在溶液中的振荡行为 ,发现巯基自组装单层膜修饰的石英晶振稳定快 ,且稳定性好 .在优化条件下 ,测得 IRS(4 9-2 0 0 0 )的滴度为 1∶ 1 50 0 .此外 ,对不同程度血吸虫感染的兔血清进行了测试 ,结果表明 ,该传感器能较好地定量区别血吸虫感染程度
A self - assembled monolayer of carboxyl - terminated mercaptopropionic acid on the surface of the piezoelectric quartz crystal gold electrode was prepared by self - assembly membrane technique. The self - assembly membrane was prepared by the reaction of 1 - ethyl - 3 - (3 - dimethylaminopropyl) carbodiimide hydrochloride and N -hydroxysuccinimide covalently immobilized 3 2 KD Schistosoma japonicum molecular antigen (Sj Ag3 2), a quartz crystal microbalance immune sensor was designed for the determination of S. japonicum antibody.Moreover, the effects of mercapto-based self-assembled monolayer and HEMA- MMA copolymer coating quartz crystal vibration behavior in solution and found that the sulfhydryl self-assembled monolayer film modified quartz crystal stabilized quickly and good stability under the optimal conditions, the measured IRS (49-2000 0) had a titer of 1: 1 50 0. In addition, different degrees of schistosoma infection in rabbit serum were tested and the results show that the sensor can be better quantitative distinction between the degree of schistosomiasis
