目的研究用甘露聚糖修饰后的甲胎蛋白(AFP)对肿瘤免疫的促进作用。方法用小鼠建立原发性肝癌模型,以AFP作为模型抗原,将重组小鼠AFP(mAFP)与氧化型的甘露聚糖相连接制成疫苗(Ox-M-mAFP),同时用未与甘露聚糖连接的mAFP作为对照,研究该疫苗在小鼠体内的肿瘤免疫治疗效果。结果Ox-M-mAFP可在小鼠体内诱导出有效的抗肿瘤免疫反应,免疫后小鼠脾内得到的CD8+T淋巴细胞显示出对特异性分泌AFP的肿瘤细胞的杀伤活性。结论本实验提供了一种有效激活机体针对自身分子免疫应答的肿瘤疫苗策略,也对原发性肝癌的非手术治疗提供了一个可能的研究方向。
Objective To study the effect of mannan-modified AFP on tumor immunity. Methods The primary hepatocarcinoma model was established in mice. AFP was used as a model antigen, and the recombinant mouse AFP (mAFP) and oxidized mannan were linked into a vaccine (Ox-M-mAFP) Glycan-linked mAFP as a control to study the tumor immunotherapeutic effect of the vaccine in mice. Results Ox-M-mAFP induced a potent anti-tumor immune response in mice. After immunization, CD8 + T lymphocytes obtained from mice spleen showed cytotoxic activity against tumor cells secreting AFP. Conclusions This experiment provides a tumor vaccine strategy that effectively activates the body against its own molecular immune response and provides a possible research direction for non-surgical treatment of primary liver cancer.
