目的:探讨表没食子儿茶素没食子酸酯(EGCG)通过Hedgehog信号抑制人胃癌MGC-803细胞增殖的机制。方法:采用不同浓度的EGCG干预人胃癌MGC-803细胞。MTT比色法观察EGCG对细胞增殖的影响;流式细胞仪检测细胞凋亡率;分光光度法检测细胞caspase-3活性;电泳法检测Smo、Gli-1蛋白表达;RT-PCR测定bcl-2,bax mRNA表达水平。结果:EGCG(10,20 mol/L)以剂量依赖性地抑制MGC-803细胞增殖,诱导其凋亡,增加caspase-3活性,抑制Smo、Gli-1蛋白表达,上调bax mRNA表达水平的同时下调bcl-2 mRNA表达水平。结论:EGCG通过Hedgehog信号抑制人胃癌MGC-803细胞增殖,诱导其凋亡。
AIM: To investigate the mechanism of epigallocatechin-3-gallate (EGCG) inhibiting the proliferation of human gastric cancer cell line MGC-803 by Hedgehog signaling. Methods: Human gastric cancer MGC-803 cells were treated with different concentrations of EGCG. MTT assay was used to observe the effect of EGCG on cell proliferation; flow cytometry was used to detect apoptosis rate; spectrophotometry was used to detect the activity of caspase-3; electrophoresis method was used to detect the expression of Smo and Gli-1 protein; bcl-2 , bax mRNA expression level. Results: EGCG (10, 20 mol / L) inhibited the proliferation of MGC-803 cells in a dose-dependent manner, induced its apoptosis, increased the activity of caspase-3, inhibited the expression of Smo and Gli-1 and upregulated the expression of bax mRNA Downregulate the expression of bcl-2 mRNA. Conclusion: EGCG can inhibit the proliferation and induce the apoptosis of human gastric cancer MGC-803 cells by Hedgehog signaling.
